Piperlongumine, or piplartine, is a natural alkaloid substance extracted from the fruit of rots of the long pepper plant (Piper longum Linn.). The plant grows in the wild tropical rain forests of India, Indonesia, Malaysia, Nepal, Philippines, and Sri Lanka, and it has been considered one of the main components in the Ayurvedic medicine with important potential to support the human body when dealing with various health challenges.
During the recent years, Piperlongumine has gain important scientific attention in many areas of health, with predominance in oncology and longevity space.
Indeed, in oncology it has been researched in relation to its potential to selectively targets tumor cells by generating cellular ROS, and a good number of key enzymes have been identified as targets of piperlongumine, such as NF-κB, STAT3, and TXNRD1.
In the longevity area, Piperlongumine stands out as it has been found to have relevant senolytic properties. It has been shown to induce apoptosis in senescent cells on its own and to be synergic with other substances.
The main mechanism that seems to stay at the basis of Piperlongumine’s relevance in oncology and longevity area, is related to its activity towards inhibiting intracellular mechanisms responsible for antioxidant productions. As such, cells that have a higher sensitivity to reactive oxygen species such as senescent cells may be affected by Piperlongumine
